Description

Clomiphene citrate, an oral medication, belongs to the selective estrogen receptor modulator (SERM) class. It functions as an antiestrogen within the triphenylethylene group, possessing both agonistic and antagonistic properties. While it blocks estrogen action by binding to receptors in specific tissues, it activates estrogen receptors in bone tissue, providing the beneficial effect of preventing osteoporosis. In males, Clomiphene acts as an antiestrogen, inhibiting receptors throughout the body. It also counteracts the negative feedback of estrogens on the hypothalamus, stimulating the production of gonadotropin-releasing hormone (GnRH), luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone, and sperm in the testicles.

Research conducted in the late 70s at the University of Ghent in Belgium compared the effects of Tamoxifen and Clomid on testosterone levels. The study found that a 10-day course of Tamoxifen at a dosage of 20mg per day increased serum testosterone levels by 142% from baseline, a result comparable to that achieved with 150mg of Clomid daily.

Differences with clomiphene

Tamoxifen does not affect blood estrogen levels. Its mechanism of action involves blocking estrogen receptors, including those in the pituitary gland. Clomiphene, on the other hand, exhibits more selectivity towards the pituitary gland, while Tamoxifen’s binding occurs more evenly throughout the body. Tamoxifen is therefore a preferred solution for estrogen-induced gynecomastia in men.

Clomiphene acts as an estrogen receptor antagonist, reversing the inhibitory effects of estrogens on the hypothalamus and pituitary gland. It aids in the rapid restoration of their functions. Tamoxifen, on the other hand, blocks receptors in almost all tissues and assists the body in restoring its own testosterone production. It should be noted that Tamoxifen is less effective in blocking estrogen receptors in the pituitary gland compared to Clomiphene. Neither Clomiphene nor Tamoxifen affect estrogen metabolism, allowing the liver enzymes to continue metabolizing estrogens without accumulation in the body.

Warnings

Tamoxifen increases the number of progesterone receptors, so it should not be used when a cycle includes progestin drugs such as nandrolone or trenbolone. In such cases, Clomiphene is the preferred choice.

Dosage

The typical recommended dosage for Tamoxifen is 10-40 mg per day, taken once or twice daily.
